Transformed African Green Monkey Kidney Fibroblast Cells (COS-7 Line)

In a double immmunofluorescence experiment, fixed and permeabilized COS-7 cells were treated with a cocktail of mouse anti-fibrillarin and goat anti-giantin primary antibodies, targeting the nucleoli and Golgi complex, respectively. The organelles were visualized with secondary antibody Fab fragments conjugated to Alexa Fluor 488 and 568 (pseudocolored yellow and red). Nuclei were counterstained with Hoechst 33342. Images were recorded in grayscale with a Hamamatsu ORCA-AG camera system coupled to an Olympus BX-51 microscope equipped with bandpass emission fluorescence filter optical blocks provided by Semrock. During the processing stage, individual image channels were pseudocolored with RGB values corresponding to each of the fluorophore emission spectral profiles.
